Is there the notion to compare T cells of known antigen specificity (by tetramers, transgenes), or just analyzing cells of unknown specificity?

T cells can be found in every possible body location, and in myriads of immunological responses. And T cell reponses vary according to dose, context and timing of exposure to antigen and other triggers. How could these all be covered?
